A practical reference on freeze-thaw: what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.

Purity and identity are separate measurements and are often confused. Reverse-phase high-performance liquid chromatography, usually with ultraviolet detection near 214 nanometres, reports the share of total peak area belonging to the target compound. Mass spectrometry by electrospray or matrix-assisted laser desorption then checks whether the observed mass matches the expected sequence. Neither measurement alone shows that a vial holds the intended peptide. Peptide content, meaning the fraction of vial mass that is genuine peptide rather than counter-ion, water or residual acid, is reported separately and is frequently lower than the stated purity figure.
The regulatory position is broadly consistent across major jurisdictions: no thymosin beta-4 fragment is an approved medicine, and laboratory material is commonly labelled as not intended for human consumption. Anti-doping rules in sport list thymosin beta-4 and its fragments among prohibited peptide hormones. Because these products travel through research-chemical channels rather than pharmaceutical supply chains, quality varies considerably between vendors. Independent testing of identity, purity and sterility is the only dependable check, and a certificate of analysis describes one batch rather than a supplier's whole catalogue.
Lyophilized peptide powder is normally held desiccated at −20 °C, with −80 °C used for longer storage periods. Allowing a sealed vial to reach room temperature before opening is standard practice, because condensation forming on cold powder introduces moisture. Once dissolved, solutions are typically kept cold and shielded from light. Repeated freeze-thaw cycles are avoided because they encourage aggregation and gradual loss of material. These conventions are general to synthetic peptides rather than unique to any one sequence.
Identity and purity are assessed with a small set of standard techniques. Reverse-phase high-performance liquid chromatography gives a purity estimate from peak area, usually recorded at 214 or 220 nanometers, where the peptide bond absorbs. Mass spectrometry confirms the expected molecular mass and can reveal truncated or oxidized species. Amino acid analysis or tandem mass spectrometry sequencing can verify the sequence itself. Additional quality attributes include water content, residual trifluoroacetic acid carried over from purification, and endotoxin where the material is intended for biological work.
The compound is most often distributed as a lyophilized powder, appearing white to off-white and forming a loose cake or fluffy solid. It is hygroscopic to some degree, so brief exposure to humid air can add water weight and complicate weighing. The peptide dissolves readily in water and in neutral aqueous buffers, and aqueous solubility is generally described as high, well above the concentrations used in typical assays. Some polar organic solvents are also usable, which matters when a concentrated stock is prepared before dilution into buffer.

Interest in the fragment grew during the 1990s and 2000s, when it moved from laboratory work into sports and supplement markets. Anti-doping bodies added thymosin beta-4 fragments to prohibited lists, and a small number of adverse analytical findings have been reported in competition testing. Published controlled human trials remain scarce. Most mechanistic evidence comes from cell culture and animal models, and those studies examine endpoints such as cell migration, wound closure and inflammation markers. That evidence supports research interest but does not establish clinical benefit, and broad regenerative claims should be read as unverified.
TB-500 is a short synthetic peptide sold under a trade name rather than a systematic chemical name. Suppliers usually describe it as a fragment of thymosin beta-4 and ship it as a lyophilised powder intended for laboratory use. Because the label is commercial, the exact sequence attributed to it is not fully consistent across catalogues, and some listings present a seven-residue peptide while others describe related fragments of similar length. It is not an approved medicine in any major jurisdiction, and it is handled as a research chemical.
Thymosin beta-4 contains 43 amino acids and has a reported molecular mass near 4963 Da. The short fragment most often associated with the TB-500 label, an acetylated chain beginning LKKTETQ, has a reported mass near 889 Da, so the two are easily separated in analytical work. Mass spectrometry and amino acid analysis can confirm which material is present in a given sample. Statements treating TB-500 and thymosin beta-4 as interchangeable are therefore imprecise, even though the two appear together in much of the same literature.
Interest in the compound comes largely from studies of the parent protein, which participates in actin sequestration, cell migration and tissue repair processes. Whether a short fragment reproduces those activities is a separate question that remains open in the published record. Many summaries describe mechanisms by analogy to thymosin beta-4 rather than from direct measurements on the fragment. Claims about activity should be treated as provisional unless a cited study specifies the exact peptide, its purity and the assay used.
TB-500 is a laboratory label applied to a short synthetic peptide that is widely described as a fragment of thymosin beta-4, an actin-binding protein present in most mammalian cells. Suppliers and review articles usually present TB-500 as the N-terminal region of that protein, but the exact sequence attached to the name is not consistent across sources. Some product descriptions list a seven-residue chain; others use the label loosely for the parent protein itself. Because of that variation, any technical discussion of TB-500 needs to state which sequence is meant.
Detection in biological samples relies on mass spectrometry, typically liquid chromatography coupled to tandem mass spectrometry after peptide extraction and enrichment. Intact peptides can also be confirmed by high-resolution mass measurement together with fragmentation data. Detection windows in urine are short because the peptide is degraded by proteases and cleared quickly, and concentrations are low. Many jurisdictions treat the compound as a prohibited substance in sport, grouped with peptide hormones and related factors, while it is not an approved therapeutic product. Identity and purity statements therefore rest on certificates of analysis, ideally issued by an independent laboratory.
Material is normally supplied as a lyophilised powder in a sealed vial. The powder is hygroscopic, so exposure to humid air leads to water uptake, caking and gradual loss of the fluffy texture that indicates a good freeze-dry. Vials are best kept sealed with desiccant, protected from light and stored cold. Letting a cold vial warm to room temperature before opening reduces condensation on the contents. Purity is normally reported from a chromatographic run, and that figure applies to the batch as tested rather than to the vial after repeated opening.
Once in solution, short peptides are generally less stable than the dry powder, and repeated freeze-thaw cycles are a common cause of loss. Laboratory guidance usually calls for aliquoting on first dissolution and storing aliquots at -20 °C or below, away from light. Adsorption to plastic and glass surfaces can lower measured concentration, particularly at low concentrations, so container material and buffer choice can affect results. Visible cloudiness, colour change or unexpected precipitate is a signal to re-check the material.
Purity is normally assessed by reversed-phase HPLC, with the main peak reported as a percentage of total peak area, while identity is confirmed by mass spectrometry. Electrospray and MALDI-TOF instruments are both used, and the observed mass is compared with the value calculated from the stated sequence. Ion-exchange or size-exclusion methods appear where aggregation or charge variants are of interest. Water content, counter-ion content and residual trifluoroacetate from purification are separate variables that can shift the measured mass and should be weighed when reading a certificate of analysis.
A 2007 study found that carriers of at least one C allele (variant CC or CT) are 7.72 times more likely than non-carriers to achieve remission after 4 weeks of treatment with amitriptyline, citalopram, paroxetine or venlafaxine (all P-gp substrates). The study included patients with mood disorders other than major depression, such as bipolar II; the ratio is 9.4 if these other disorders are excluded. At the 6-week mark, 75% of C-carriers had remitted, compared to only 38% of non-carriers.

=== 2022 medication shortage === In November 2022, Australia's medical regulatory agency, the Therapeutic Goods Administration (TGA) reported that there was a global shortage of the diabetes medication Ozempic. According to the TGA, the rise in demand was caused by an increase in off-label prescription of the drug for weight loss purposes. In December 2022, with the United States experiencing a shortage as well, it was reported that the huge increase in demand for the medicine was caused by a weight loss trend on TikTok, where videos about the drug exceeded 360 million views. Wegovy, a drug that has been specifically approved for treating obesity, also became popular on the platform after Elon Musk credited it for helping him lose weight.
=== Cholecystitis === Total parenteral nutrition increases the risk of acute cholecystitis due to complete disuse of the gastrointestinal tract, which may result in bile stasis in the gallbladder. Other potential hepatobiliary dysfunctions include steatosis, steatohepatitis, cholestasis, and cholelithiasis. Six percent of patients on TPN longer than three weeks and 100% of patients on TPN longer than 13 weeks develop biliary sludge. The formation of sludge is the result of stasis due to lack of enteric stimulation and is not due to changes in bile composition. Gallbladder sludge disappears after four weeks of a normal oral diet. Administration of exogenous cholecystokinin (CCK) or stimulation of endogenous CCK by a periodic pulse of large amounts of amino acids has been shown to help prevent sludge formation. These therapies are not routinely recommended. Such complications are suggested to be the main reason for mortality in people requiring long-term total parenteral nutrition, such as in short bowel syndrome. In newborn infants with short bowel syndrome with less than 10% of expected intestinal length, thereby being dependent upon total parenteral nutrition, five-year survival is approximately 20%.

This is a list of scandals or controversies whose names include a -gate suffix, by analogy with the Watergate scandal in the 1970s, as well as other incidents to which the suffix has (often facetiously) been applied. This list also includes controversies that are widely referred to with a -gate suffix, but may be referred to by another more common name (such as the New Orleans Saints bounty scandal, known as "Bountygate"). Use of the -gate suffix has spread beyond American English to many other countries and languages.
=== Pasteurization === Inactivation of viruses by means of pasteurization can be very effective if the proteins that you are trying to protect are more thermally resistant than the viral impurities with which they are in solution. Some of the more prominent advantages of these types of processes are that they require simple equipment and they are effective for both enveloped and non-enveloped viruses. Because pasteurization involves increasing the temperature of solution to a value that will sufficiently denature the virus, it does not matter whether the virus has an envelope or not because the envelope alone cannot protect the virus from such high temperatures. However, there are some proteins which have been found to act as thermal stabilizers for viruses. Of course, if the target protein is not heat-resistant, using this technique could denature that target protein as well as the viral impurity. Typical incubation lasts for 10 hours and is performed at 60 °C.
Valve has released two versions of a game controller aimed to be used for Steam games, the Steam Controller. The first model was released in November 2015, alongside the original Steam Machine concept, while the second was released in May 2026 ahead of Valve's dedciated Steam Machine model and Steam Frame. Among other features, both generations of the controller use two small trackpads with haptic feedback which can be used either as traditional controller actions like a joystick, but also like a computer mouse input to control many personal computer games that lack traditional controller support.
Meristematic cells give rise to various organs of the plant and keep the plant growing. There are two types of meristematic tissues 1) Apical Meristem 2) Lateral Meristem. The Apical Meristem is of two types; the shoot apical meristem (SAM) gives rise to organs like the leaves and flowers, while the root apical meristem (RAM) provides the meristematic cells for the future root growth. SAM and RAM cells divide rapidly and are considered indeterminate, in that they do not possess any defined end status. In that sense, meristematic cells are frequently compared to animal stem cells, which have an analogous behavior and function. Within plants, SAM cells play a major role in the overall growth and development, this is due to the fact that all cells making up the major parts of the plant come from the shoot apical meristem (SAM). There are three different important areas found within the SAM and they include the central zone, the peripheral zone, and the rib meristem. Each of these areas play an important role in the production of new stem cells within the SAM. All SAMs are usually dome shaped and have structures that are layered and are described as the tunica and corpus. CLV3 plays an important role in regulating the production of stem cells within the Central Zone region of the (SAM), this is also true for the cell promoting WUSCHEL (WUS) gene. The combination of these two genes regulates stem cell production by WUS negatively or positively regulating the production of stem cells by controlling the CLV3 gene.
=== Pediatrics === Fever is often viewed with greater concern by parents and healthcare professionals than might be deserved, a phenomenon known as fever phobia, which is based in both caregiver's and parents' misconceptions about fever in children. Among them, many parents incorrectly believe that fever is a disease rather than a medical sign, that even low fevers are harmful, and that any temperature even briefly or slightly above the oversimplified "normal" number marked on a thermometer is a clinically significant fever. They are also afraid of harmless side effects like febrile seizures and dramatically overestimate the likelihood of permanent damage from typical fevers. The underlying problem, according to professor of pediatrics Barton D. Schmitt, is that "as parents we tend to suspect that our children's brains may melt." As a result of these misconceptions parents are anxious, give the child fever-reducing medicine when the temperature is technically normal or only slightly elevated, and interfere with the child's sleep to give the child more medicine.

Desiccated storage at −20 °C is conventional, with −80 °C for extended periods. Vials should be warmed to room temperature before opening to prevent condensation on the powder.
It usually reports batch-specific results for purity by chromatography, identity by mass spectrometry, and sometimes peptide content and residual solvents. It applies only to the batch tested, not to a supplier's wider stock.
Purity describes the proportion of the chromatographic signal from the target compound, while peptide content measures how much of the vial's physical mass is peptide. Counter-ions, water and residual acid make up the remainder, so content values are commonly lower.
Short transit at ambient temperature is generally tolerated, but long-term storage at room temperature is not recommended. Heat, moisture, and light all accelerate degradation. Cold, dry, dark storage is the conventional choice.
